What is N-Acetyl Semax?
N-Acetyl Semax Amidate is an analog of the neuropeptide Semax, itself a synthetic analog of ACTH(4-10). It is characterized by an N-terminal acetylation and C-terminal amidation, modifications intended to increase its metabolic stability and permeability across the blood-brain barrier, potentially leading to a longer half-life and enhanced nootropic and neuroprotective effects compared to the parent compound Semax. Research suggests its mechanisms involve modulation of neurotrophic factors like BDNF and NGF, influencing neuronal plasticity and survival.
Key Benefits
- Enhanced cognitive function
- Memory improvement
- Neuroprotection
- Greater stability than Semax
- Focus and mental clarity
Mechanism of Action
- Similar to Semax with improved stability
- BDNF and NGF pathway modulation
- Crosses blood-brain barrier
Best For
- Cognitive performance research
- Neuroprotection

Research Indications
Improved Focus and Attention
Research indicates a potential for enhanced concentration and sustained attention, particularly in mentally demanding tasks.
Memory Optimization
Studies suggest benefits in both short-term recall and the consolidation of long-term memories.
Research Protocols
| Goal | Dose | Frequency | Route |
|---|---|---|---|
| General Cognitive Enhancement | 300 mcg | Once daily | Subcutaneous injection or Nasal spray |
| Enhanced Focus for Demanding Tasks | 500 mcg | Once daily, 1-2 hours before task | Subcutaneous injection or Nasal spray |
| Neuroprotection Study | 300-600 mcg | Once daily | Subcutaneous injection or Nasal spray |
| Memory Improvement Protocol | 400 mcg | Once daily | Subcutaneous injection or Nasal spray |
| Exploring Mood Regulation | 300 mcg | Once daily | Subcutaneous injection or Nasal spray |
Nasal administration is also a commonly discussed route, with similar dosing strategies. When using nasal spray, ensure proper atomization for optimal absorption.
Peptide Interactions
- Synergistic
Piracetam and other racetams
Often discussed for synergistic cognitive benefits, potentially enhancing acetylcholine system modulation.
- Synergistic
Noopept
Commonly combined with N-Acetyl Semax Amidate in research for enhanced neurotrophic and cognitive effects.
- Monitor Combination
Stimulants (e.g., Caffeine, Modafinil)
Potential for increased stimulation or restlessness; close monitoring of subject response is advised.
- Use Caution
Sedatives/Anxiolytics
Potential for altered effects or reduced efficacy of either compound; avoid concurrent administration unless specifically studied.
- Avoid
Alcohol
Alcohol can negate the cognitive benefits and potentially exacerbate side effects; generally advised to avoid.
- Use Caution
Antidepressants (SSRIs, SNRIs)
Potential for altered neurotransmitter balance; combination should only be explored under strict research protocols and supervision.
How to Reconstitute
- 1Gather N-Acetyl Semax Amidate vial, bacteriostatic water, and sterile syringes.
- 2Clean the rubber stopper of both vials with alcohol wipes.
- 3Draw the desired amount of bacteriostatic water (e.g., 1-2 mL) into a sterile syringe.
- 4Slowly inject the bacteriostatic water into the N-Acetyl Semax Amidate vial, directing it down the side of the glass.
- 5Gently swirl the vial to dissolve the powder. Do not shake.
- 6Once fully dissolved, store the reconstituted solution in the refrigerator.
Use bacteriostatic water for reconstitution. Do not shake vigorously; gently swirl to mix.
What to Expect
- Weeks 1-2: Initial subtle improvements in focus and mental clarity may be noticed.
- Weeks 2-4: More pronounced cognitive benefits, such as improved memory recall and learning capacity, are commonly discussed.
- After 4 weeks: Sustained improvements in mood and reduced mental fatigue may be reported in research contexts.
- Post-cycle: A return to baseline cognitive function is generally expected after cessation.
- Individual responses can vary significantly based on genetics and other factors.
Side Effects & Safety
