What is Epithalon?

Epithalon (Ala-Glu-Asp-Gly) is a synthetic tetrapeptide modeled on epithalamin, a crude polypeptide extract of bovine pineal glands. Research suggests its primary mechanism involves activation of telomerase, the enzyme that maintains and elongates telomeres, the protective caps on chromosome ends. Additional research protocols investigate antioxidant properties, pineal gland modulation, melatonin secretion and circadian rhythm effects. An important dosing distinction is often missed. The widely repeated 5-10 mg per day figure traces back to studies using Epithalamin, the crude extract, not the purified synthetic peptide. Epithalamin contains a mixture of components of which only a fraction is the active peptide, so a large milligram dose was needed. Primate comparisons suggest a potency differential on the order of several hundred fold, and a Drosophila antioxidant model suggested roughly 1000-fold, with Epithalon outperforming Epithalamin despite far lower dosing. Allometric scaling of the effective mouse treatment-day dose (about 1 microgram per mouse, 5 consecutive days per month) gives a human equivalent of roughly 190-230 mcg per treatment day for a 70 kg adult. In vitro data also show a non-monotonic response, with peak telomere elongation around 0.2 mcg/mL and weaker effects at higher concentrations. Taken together, current analysis places Epithalon in the low microgram range rather than the milligram range.

The longevity peptide, and the one with the biggest dosing mix-up in the whole space. One study of 266 volunteers reported 1.6-1.8 times reduced mortality versus controls - but that study used Epithalamin, the crude bovine extract, not the synthetic tetrapeptide everyone is actually buying. That is where the 5-10 mg advice came from, and it very likely does not apply here. Scaled from the animal work, Epithalon lands in the 100-300 mcg range per dosing day. If you have been running 10 mg, you are probably not getting a stronger effect - just an expensive one. The telomerase mechanism is genuinely fascinating; the dosing convention needs to catch up.

How to Reconstitute

  1. 1Gather Epithalon vial, bacteriostatic water (BW), sterile syringe (e.g., 1mL), and alcohol wipes.
  2. 2Carefully remove the protective cap from the Epithalon vial and wipe the rubber stopper with an alcohol wipe.
  3. 3Draw the desired amount of bacteriostatic water into the syringe. A common ratio is 1ml of BW per 5mg or 10mg vial.
  4. 4Insert the syringe needle into the Epithalon vial and slowly inject the bacteriostatic water, allowing it to run down the inside wall of the vial.
  5. 5Do not inject directly onto the lyophilized powder. Avoid shaking the vial; gently swirl until the powder is fully dissolved.
  6. 6Once dissolved, the solution is ready for administration. Store according to guidelines.
